Development and validation of new SSR markers from expressed regions in the garlic genome Scientia Agricola
Ipek,Meryem; Sahin,Nihan; Ipek,Ahmet; Cansev,Asuman; Simon,Philipp W..
Only a limited number of simple sequence repeat (SSR) markers is available for the genome of garlic (Allium sativum L.) despite the fact that SSR markers have become one of the most preferred DNA marker systems. To develop new SSR markers for the garlic genome, garlic expressed sequence tags (ESTs) at the publicly available GarlicEST database were screened for SSR motifs and a total of 132 SSR motifs were identified. Primer pairs were designed for 50 SSR motifs and 24 of these primer pairs were selected as SSR markers based on their consistent amplification patterns and polymorphisms. In addition, two SSR markers were developed from the sequences of garlic cDNA-AFLP fragments. The use of 26 EST-SSR markers for the assessment of genetic relationship was...

Estimation of genetic and environmental relationships between milk yield and different measures of mastitis and hyperkeratosis in Holstein cows Animal Sciences
Chegini, Arash; Hossein-Zadeh, Navid Ghavi; Hosseini-Moghadam, Hossein; Shadparvar, Abdol Ahad.
In this research, records of milk yield, mastitis occurrence and teats hyperkeratosis of 453 Holstein cows from a large herd with calving dates from December 2012 to December 2014 was used to estimate genetic and environmental correlations between milk yield, mastitis and hyperkeratosis. The highest and the lowest number of mastitis occurrence were in winter (January to March) and spring (April to June), respectively. Also, the highest and the lowest averages of days in milk at the time of mastitis occurrence were observed in winter and spring, respectively (162.53 vs. 29.67 days). The total number of mastitis occurrence was 151 times and in average a sick cow was infected 1.649 times. Obtained results showed low heritabilities for measures of mastitis...

Evaluation of Bayesian models for analysis of crude protein requirement for pigs of Brazilian Piau breed Scientia Agricola
Silva,Hugo Teixeira; Silva,Fabyano Fonseca e; Ferreira,Aloízio Soares; Veroneze,Renata; Lopes,Paulo Sávio.
ABSTRACT: We evaluated the inclusion of information on genetic relationship into the analysis of crude protein requirement in diets for pigs of Brazilian Piau breed, using Bayesian inference. The animals were assigned to treatments in a completely randomized design in factorial scheme 4 × 2 (crude protein levels × sex) with 12 repetitions per treatment. The evaluations were carried out in the initial, growing and finishing phases, and after slaughter. The traits evaluated were feed conversion (FC), backfat thickness (BF), daily weight gain (DWG), daily feed intake (DFI) and some carcass cuts. Three models were considered to evaluate the inclusion of information on genetic relationship into the analysis: Model I, a simple linear model; Model II, the same...

Genetic relationships among species of the genus Bothrops based on RAPD markers BABT
Grazziotin,Felipe; Echeverrigaray,Sergio.
The genetic relationships among 11 species of Bothrops found in Brazil were determined using random amplified fragment (RAPD) information. A total of 239 amplified bands were scored using 20 aleatory primers. Most of the bands (88.7%) were polymorphic. Phylogenetic analysis of the data determined three similarity groups within Bothrops: Group I- B. alternatus, B. neuwiedi, B. cotiara, and B. jararacusu; Group II- B. insularis, B. jararaca, and B. erythromelas; and Group III- B. moojeni, B. leucurus, and B. atrox. These groups coincided with those obtained by other methods, indicating that RAPD's could be a useful tool for the evaluation of genetic relationships at the interspecific level.

Genetic relationships between Chinese, Japanese, and Brazilian soybean gene pools revealed by simple sequence repeat (SSR) markers Genet. Mol. Biol.
Yamanaka,Naoki; Sato,Hiroyuki; Yang,Zhenyu; Xu,Dong He; Catelli,Lizandra Lucy; Binneck,Eliseu; Arias,Carlos Alberto Arrabal; Abdelnoor,Ricardo Vilela; Nepomuceno,Alexandre Lima.
An understanding of the relationship of geographically different soybean gene pools, based on selectively neutral DNA markers would be useful for the selection of divergent parental cultivars for use in breeding. We assessed the relationships of 194 Chinese, 59 Japanese, and 19 Brazilian soybean cultivars (n = 272) using 12 simple sequence repeat (SSR) markers. Quantification Theory III and clustering analyses showed that the Chinese and Japanese cultivars were genetically quite distant to each other but not independent, while Brazilian cultivars were distantly related to the cultivars from the other two countries and formed a cluster that was distant from the other two gene pool clusters. Our results indicated that the Brazilian soybean gene pool is...

Heterologous microsatellite primer pairs informative for the whole genus Arachis Genet. Mol. Biol.
Hoshino,Andrea Akemi; Bravo,Juliana Pereira; Angelici,Carla M.L.C.D.; Barbosa,Andréa V. Gobbi; Lopes,Catalina Romero; Gimenes,Marcos A..
The genus Arachis currently comprises 69 described species, some of which have potential and real value as human and animal foods. These Arachis species have been collected and maintained in germplasm banks to provide material that can be used as sources of genes in breeding programs and for the selection of new cultivars. One of the principal objectives of germplasm conservation is the evaluation of genetic variability, which is best conducted using molecular markers. We investigated the use of heterologous primers to amplify microsatellite loci that could be used to evaluate genetic variability in Arachis germplasm. Fifteen microsatellite primer pairs were tested in 76 accessions of 34 species from the nine Arachis sections. The data indicated that...

MORPHOLOGICAL TRAITS AS TOOL TO VERIFY GENETIC VARIABILITY OF INTERSPECIFIC DRAGON FRUIT HYBRIDS Rev. Bras. Frutic.
SILVA,ADRIANA DE CASTRO CORREIA DA; SABIÃO,RAFAEL ROVERI; CHIAMOLERA,FERNANDO MARCELO; SEGANTINI,DANIELA MOTA; MARTINS,ANTONIO BALDO GERALDO.
ABSTRACT With recent cultivation in Brazil, the dragon fruit still have lacks related to the selection of promising materials and there are not yet commercial varieties in the country. Therefore, manual pollination cross were held in 2010, between plants of H. undatus x H. polyrhizus and H. undatus x H. setaceus. Aiming evaluate the genetic diversity of the progenies for future use in breeding program, based on cladodes characteristics, six characters were assessed: length and diameter of stem, distance between areoles, arch height, number and size of spines/areole. From the Euclidean distance matrix analysis was constructed a dendrogram by the UPGMA method. There was great variability among hybrids, and eight of them have shown promise for use in...

Relationships among six herbal species (Curcuma) assessed by four isozymes Phyton
Deng,JB; Ding,CB; Zhang,L; Zhou,YH; Yang,RW.
Four isozymes, superoxide dismutase (SOD), polyphenol oxidase (PPO), malate dehydrogenase (MDH) and cytochrome oxidase (COD) were studied for identification of six herbal species (Curcuma L.). All the 37 study specimens produced a total of 168 polymorphism isozyme bands. The genetic distance coefficients (GS) varied from 0.08 to 0.54. The dendrogram, obtained according to the polymorphism isozyme bands by the UPGMA method with the software NTSYS-pc2.1, contributed to improve the resolution of phylogeny. From the dendrogram, it was possible to differentiate between the wild and cultivated specimens of C. longa, and within C. sichuanensis species.
